摘  要:目的 探讨乙酰肝素酶反义寡核苷酸(AS-ODN)对人胃癌细胞株SGC7901表达碱性成纤维细胞生长因子(basic fibrob-last growth factor,bFGF)的影响。方法 AS-ODN和乙酰肝素酶mRNA起始密码子区互补,无义寡核苷酸(NS-ODN)为对照组,用阳离子脂质体包裹ODNs并转入SGC7901细胞;转染后48小时提取细胞总RNA,半定量RT-PCR法检测乙酰肝素酶基因的含量,免疫细胞化学法检测bFGF的表达。结果 AS-ODN处理后SGC7901表达乙酰肝素酶mRNA下降;处理前bFGF的表达率为72.23%,不同浓度(0.1、0.2、0.4、0.8μmol/L)AS-ODN处理后bFGF的表达率下降程度不同(分别为57.15%、51.11%、42.36%和40.25%)。结论 和乙酰肝素酶mRNA起始密码子区互补的AS-ODN对SGC7901表达bFGF有明显影响,且呈剂量相关性。Objective To evaluate the effect of heparanase antisense oligodeoxynucleotide(AS-OND) on the expression of basic fibroblast growth factor(bFGF) in human gastric carcinoma cell line SGC7901 .Metheds The ODNs were embedded in cationic liposome lipofectin and transfected into SGC7901 cells. The total RNAs were extracted from the cells 48 hours after transfection and the semi-quantitative RT-PCR were performed to evaluate the heparanase gene. The change of bFGF was observed by using immunocytochemistry. Results The heparanase gene expression and the expression of bFGF of SGC7901 cells treated with AS-ODN of different final concentrations were significantly decreased compared with that of the controls ( P < 0 01) . The expression rates were 72.23% , 57.15%, 51.11%, 42.36% and 40.25% before and after treated with AS-ODN of the final concentrations of 0.1 μmol/L,0.2 /μmol/L,0.4μmol/L and 0.8μmol/L, respectively. Conclusion Heparanase AS-ODN complementary to the start codon region of heparanase mRNA has a significant effect on the expression of bFGF in human gastric carcinoma cell line SGC7901 in a dose-depended manner.
