摘  要:γ-氨基丁酸(GABA)是一种具有生理活性的氨基酸,用生物发酵方法合成的GABA需要进一 步的分离和纯化.针对发酵液成分的复杂性,采用高速离心进行初步的分离,然后运用超滤和稀释 过滤相结合的方法,在去除大分子的同时,尽量提高GABA的回收率.用分子切割量为50000 D的 PES膜组件进行试验,研究了各操作参数的影响,得到优化参数:温度低于40℃,压差0.2 MPa,循 环流速0.5 m/s,pH=5.因超滤后GABA的回收率较低,故采用稀释过滤,以提高回收率.在两次间 歇式稀释过滤后,GABA的回收率由原来的50%提高到将近95%,达到较好的分离效果.超滤后的 滤液可再作进一步的纯化处理.γ-aminobutyric acid (GABA) is a bioactive amino acid. GABA produced by fermentation should be separated and purified. Due to the complexity of composition, the fermentation was at first treated by high speed centrifuge. Then the combined UF and DD processes were applied to purify GABA. The PES membrane with MWCO value of 50000 was used in the experiment. The influences of operation parameters were investigated and the optimization led to the following conditions: t < 40°C, Δp = 0.2 MPa, μ = 0.5 m/s, pH = 5. The recovery ratio of GABA in the UF was not high, so a diafiltration was necessary. After two times of discontinuous diafiltration (DD), the recovery ratio was increased from 50% to nearly 95%. The filtrate can be further purified in the next step.
