黄连素及其与BCNU联合应用治疗恶性脑肿瘤的实验研究  被引量:5

摘  要:本文对黄连素的抗脑肿瘤作用,以及与1,3-bis(2-chloroethyl)-l-nitrosourea(BC-NU)联合应用进行了实验研究。用集落生成率测定(CFE)对6个人类脑胶质瘤细胞株和大白鼠9L脑肿瘤细胞(胶质肉瘤)进行了离体研究。用姐妹染色体交换法(SCE),对9L脑肿瘤细胞进行了评价。同时,在活体实验中用集落生成率测定对脑内固体9L脑肿瘤的治疗进行了分析。在BCNU的浓度为23μm单独使用时,对6个人类脑胶质瘤细胞株可获得平均为43%的细胞杀灭作用(0.24Log Kill)。而在黄连素与23μmBCNV联合应用治疗中,则可见到平均为97%的细胞杀灭相加作用(1.55Log Kill)。在采用150μg/ml黄连素离体治疗中,9L细胞姐妹染色体相对数的增高超过底数的2.7倍。在活体实验中,黄连素与BCNU联合应用也显示出细胞毒性相加作用。Berberine was evaluated for antitumor activity against malignant brain tumors.In addition,the synergy effect of berberine and BCNU was done.In vitro the studies were performed on a series of 6 human malignant brain tumor cell lines and rat 9L brain tumor cells (gliosarcoma) by using a colony forming efficiency (CFE) assay.9L was also evaluated by a sister chromatid exchange (SCE) assay.In addition,in vivo,the treatment of intracerebral 9L solid brain tumors was analyzed by CFE assay.On the average BCNU alone at a dose of 23 uM gave a 43% cell kill (0.24 log kill).The treatment of berberine in combination with BCNU at 23 uM showed additive effects with an average of 97% cell kill (1.55 log kill).The relative number of SCEs for 9L cells was increased 2.7 times over background following in vitro treatment with 150 ug/ml of berberine.In vivo,the combination treatment of berberine and BCNU also showed additive cytotoxicity.
