摘 要:本文用透射电镜结合扫描电镜观察了转基因满江红鱼腥藻细胞形态和超微结构的变化。与对照细胞相比较,转基因满江红鱼腥藻细胞外被有粘性分泌物堆集,不如对照光滑。转化藻丝较对照藻丝短。此外,异细胞分布频率也有较大变化,5#转基因藻比对照高出25%。透射电镜观察发现转基因藻细胞外壁结构,内膜系统和内含物等成份均发生了显著变化。尤其发现大量颗粒,或围绕质膜成规则排列,或呈非规则聚积散布细胞内。这些对形态和超微结构观察的结果较好地说明了转基因藻细胞光合固氮生理生化活性的变化。We had observed the cell morphology and ultrastructure of transgenic cyanobacteria Anabaena azollae withSEM and TEM. Cells of A. azollae were transformed by method of site-directed integration of NPT Ⅱ into glnAof the cells.Compared with the smooth envelope of control cells,the transgenic cells were characterized by ashrunck wall covered with sticky excritive materia. interconneetion of the transgenic is shorter than the control one. The frequency of heterocysts in transgenic strain of No. 5 was 25%higher than that of control cells. Byobservation with TEM ,we had found remarkable variances in cell wall structure , endomembrane system andvarious kinds of involved substances.Particularly those particles distributed in cells in a way of either surround-ing the endoplasma of the transgenic cells or with a few particles aggregating together.The observed results on morphology and ultrastructure of transgenic cyanobacteria were in good accordancewith their functional variation in activities of photosynthesis and nitrogen-fixation.
