摘  要:就应用强酸性阳离子交换树脂(B)及强碱性阴离子交换树脂(G)分离提纯发酵液中L-谷氨酰胺(L-Gln)的工艺条件进行了研究,通过单因子及正交试验确定离子交换分离法分离L-Gln的较佳条件为:强酸性阳离子交换树脂分离L-Gln的上柱液pH为3.5,洗脱剂氨水的浓度为0.03mol/L,洗脱速度为树脂体积的0.65%;强碱性阴离子交换树脂分离L-Gln的上柱液的pH为8.0,洗脱剂盐酸的浓度为0.05mol/L,洗脱速度为树脂体积的1.15%;使用上述条件提取L-Gln的得率为30%左右。In this article,we studied the process conditions of isolating and purifying L-Glutamine(L-Gln) from the fermentative liquid by using strong acidic cation exchange resin(B) and alkaline anion exchange resin(G).By single-factor test and rhombic test,better conditions of isolating L-Gln with ion-exchange resin method was found:L-Gln was isolated by using resin(B) with the pH of fermentative broth before putting it into the resin column being adjusted to 3.5,NH4OH elute 0.03 mol/L and eluating speed at 0.65% of the bulk of the resin;then L-Gln was isolated through resin(G) with pH of the fermentative liquid being adjusted to 8.0,NH4OH elute 0.05 mol/L and eluating-speed at 1.150% of the bulk of the resin.Applying the above conditions,L-Gln yielded at a rate about 30%.
