GTC对DMBA诱发大鼠乳腺增生性病变抑制作用的动态观察

摘　　要：目的 :绿茶提取物GTC对用致癌剂DMBA诱发的SD大鼠乳腺增生性病变的抑制作用。方法 :170只7周龄雌性SD大鼠 ,按体重分层后随机分为4组 ,即A(空白对照组 )、B(模型组 )、C(GTC对照组 )、D(实验组 )。于实验开始日 ,B组和D组每只大鼠接受一次性DMBA油溶液灌胃 ,剂量为7mg/100g 体重 ;同时C组和D组每只大鼠接受GTC水溶液灌胃 ,剂量为80mg/只/天 ,A组和B组大鼠自由饮用自来水至20周末实验结束。于实验第4、8、12、16、20周末分批处死各组大鼠 ,取乳腺组织常规制片 ,HE染色。结果 :各时间点上 ,饮茶组 (C组和D组 )和非饮茶组 (A组和B组 )相比 ,其增生性病变较少 ;这种差异在接受致癌剂的B组和D组之间 ,具有显著性 (P<0.05 ,而在对照A组和C组之间差异无显著性 (P>0.05) ;B组和D组之间的增生性病变 (芽体 +实团 )计数的差值呈现随时间延长而增加的趋势。结论 :GTC对DMBA诱导产生的大鼠乳腺增生性病变具有一定的抑制作用且这种抑制作用呈现时间依赖性递增。Objective:To illustrate the inhibitory effect of GTC(green tea catechins)on the development of the hyperplastic lesions in rat mammary gland induced by DMBA.Methods:A total of1707week old female SD rats was divided into4groups randomly:group A(blank control),B(DMBA gavage),C(GTC gavage)and D(DMBA/GTC gavage).At the beginning of the experiment,Group B and D received a7mg/100g bodyweight intragastric dose of DMBA,at the same time,group C and D received one intragastric dose of GTC(80mg/rat/day).The GTC-gavage was administrated until the end of the the20 th week.All rats got the basic fodder and tap water freely.By the end of the4 th ,8 th ,12 th ,16 th ,and20 th ,all the rats in every group were selected randomly killed averagely for each time point,the breast tissues were removed and fixed and made HE sections routinely.Results:There are more hyperplastic lesions which include solid bodies,buds,papillary hyperplsia and mitosis in group B and D than those lesions in group A and C.Compare to group C and D which got the GTC-gavage daily,there are more hyperplastic lesions in group A and B without the GTC-gavage.Such difference between group B and D are statistically significant(P<0.05),but no significance can be found between group A and C.The difference between D and B in the hyperplastic lesions are in creasing with the time passing.Conclusion:Constant GTC-gavage can inhibit the development of the breast ductal hyperplasia induced by DMBA in SD rat to some extent and such inhibitory effect seems enhancing with the time passing.
