迷走神经阻断对哮喘诱导延髓内脏带及下丘脑室旁核和视上核Fos蛋白表达的影响

摘  要:目的:研究支气管哮喘状态下迷走神经向脑传递免疫信息的作用。方法:实验于2003-10/2004-06在解放军第一军医大学珠江医院神经外科进行。清洁级雄性豚鼠20只单纯随机分为4组:假手术+生理盐水;手术+生理盐水;假手术+卵蛋白;手术+卵蛋白。建立豚鼠哮喘模型,在致敏状态下切断或假切断颈部双侧迷走神经,术后2周用卵蛋白经呼吸道激发豚鼠哮喘发作,2h后处死动物,用免疫组织化学ABC法观察延髓内脏带(MVZ)、下丘脑室旁核(PVN)和视上核(SON)Fos蛋白的表达。结果:假手术和手术组豚鼠给予生理盐水刺激后,仅在MVZ背内侧部、中间网状带和腹外侧部、SON和PVN内发现少数浅染的Fos蛋白阳性细胞。假手术组豚鼠卵蛋白激发后,MVZ,PVN和SON内可发现大量Fos蛋白表达,分别为(498.00±134.28),(99.27±15.32),(305.10±84.68)个,手术组豚鼠卵蛋白激发后上述核团内的Fos蛋白表达量则明显减少,分别为(239.78±48.28),(62.36±19.02),(127.14±57.01)个,两者比较差异有显著性意义(P<0.05)。结论:迷走神经在支气管哮喘免疫应答中,可能是传递呼吸道免疫信息的重要途径之一。AIM: To study the role of vagus nerve in conveying immune information into the brain under the condition of bronchial asthma.METHODS: The experiment was performed in the Department of Neurosurgery, Zhujiang Hospital of First Military Medical University of Chinese PLA from October 2003 to June 2004.Twenty male guinea pigs were randomly divided into four group: sham operation and saline group, operation and saline group, sham operation and ovalbumin group, operation and ovalbumin group.The ovalbumin( OVA) sensitized and challenged guinea pigs model of asthma was adopted. Guinea pigs were undertaken either bilateral cervical vagotomy or sham operation under the condition of sensitization. Two weeks later, asthma was induced in guinea pigs with OVA via respiratory tract, and killed 2 hours later. Changes of the expression of Fos protein in the medullary visceral zone (MVZ), paraventricular nucleus of hypothalamus (PVN) and supraoptic nucleus (SON) were observed with immunohistochemical ABC staining method. RESULTS:A few of lightly stained Fos positive cells were found in the dorsomedial area, intermediate reticular zone and ventrolateral area of MVZ, SON and PVN in the sham operation and operation pigs following the normal saline stimuli. OVA challenge resulted in abundant Fos protein expression within the MVZ, PVN and SON in the sham operation groups, and the amount was 498± 134.28, 99.27± 15.32 and 305.10± 84.68,respectively. And the amount of Fos positive cells in the above- mentioned nucleus was markedly reduced to 239.78± 48.28,62.36± 19.02,127.14± 57.01 in the operation groups,respectively.There was significant difference between sham operation group and operation group(P< 0.05).CONCLUSION: The vagus nerve may be one of the important signal pathways in respiratory tract to transmit immune information into the brain during the course of immune response of bronchial asthma.
