摘  要:采用100头肥育期苏太猪,屠宰取背最长肌样品。测定各样品肌内脂肪(intramuscular fat,IMF)含量,并根据IMF含量将样品分成高、中、低3个组, 每组取肌内脂肪含量位于中间的20个个体为研究对象,共计60个个体,各组IMF百分含量(%)分别为(6.00±1.14)、(9.46±0.94)和(13.77±2.12)。采用RT-PCR方法测定各样品中脂肪酸合成酶(fatty acid synthase,FAS)和激素敏感脂酶(hormone-sensitive lipase,HSL)mRNA水平,并分析其与肌内脂肪含量之间的相关性,以揭示FAS和HSL的表达与肌内脂肪沉积的关系。结果表明,FAS或HSL基因表达水平与肌内脂肪含量之间并无显著的相关(P >0.05), FAS与HSL mRNA的比值与肌内脂肪含量之间存在着显著的正相关(R =0.464,P <0.01,P =60)。不同组别之间,随着肌内脂肪含量的增加,FAS与HSL比值有升高的趋势。推测FAS和HSL在猪背最长肌肌内脂肪的沉积中发挥着作用。A hundred finishing Sutai pigs were slaughtered and longissimus dorsi (LD) muscle samples were taken for determining the intramuscular fat (IMF) contents. According to IMF contents, 60 samples were equally divided into 3 groups, containing high, median and low IMF contents. The average IMF percentage (%) of three groups were (6.00±1.14), (9.46±0.94) and (13.77±2.12), respectively. By means of semi-quantitative RT-PCR , the mRNA level of fatty acid synthase (FAS) and hormone-sensitive lipase (HSL) were detected. The correlation between FAS/HSL mRNA expression and IMF contents was analyzed to reveal the possible roles of FAS and HSL in IMF deposition in LD muscle. The result showed that the mRNA level of FAS or HSL had no significant relation with IMF content per se. However, the ratio of FAS to HSL was found to be positively correlated with IMF content (R =0.464,P <0.01,n =60). The group with high IMF content exhibited significantly higher ratio of FAS/HSL expression. It is suggested that FAS and HSL are involved in the deposition of intramuscular fat in LD muscle of pigs.
