染料木黄酮对顺铂诱导肝癌SMMC-7721细胞凋亡的增敏作用

摘　　要：目的: 探讨染料木黄酮增强顺铂对人肝癌SMMC 7721细胞生长抑制、诱导凋亡及NF κB表达的影响.方法: 采用MTT法、透射电镜、激光共聚焦显微镜、流式细胞仪和免疫组化技术研究染料木黄酮,顺铂,染料木黄酮与顺铂联合3组药物处理肝癌细胞后生长抑制、形态学变化、诱导凋亡和对NF κB表达的影响.结果: MTT法证实联合组对细胞抑制作用最强,作用24 , 48h后(A570nm: 0 497±0 030→0 295±0 098, 0 791±0 042→0 312±0 019,P<0 01 ).镜下可见细胞出现典型凋亡细胞形态学改变.流式细胞仪检测凋亡以联合组最强,细胞周期阻滞于G2 /M期.免疫组化技术检测顺铂作用后NF κB表达( 64 9±5 4 )%,联合组作用后( 23 4±2 1)% (P<0 01 ).结论: 染料木黄酮通过抑制激活NF κB表达,增强顺铂诱导肝癌SMMC 7721细胞凋亡.AIM: To investigate the inhibitory effects of genistein combined with cisplatin on the anti-proliferation,apoptotic induction and expression of NF-κB in human hepatocellular cancer cell SMMC-7721.METHODS: After the respective treatment with genistein,cisplatin and genistein combined with cisplatin,the cell viability was examined by methyl thiazolyl tetrazolium assay.The morphology of cells was observed respectively under transmission electron microscope and laser scanning confocal microscope.The flow cytometry was used to detect the apoptosis rates and cell cycle distribution.The expressions of NF-κB were determined by the immunocytochemical method.RESULTS: MTT showed that the inhibitory effect in the co-treatment group was more obvious after exposure to drugs for 24 h and 48 h (A 570 nm : 0.497±0.030→0.295±0.098,0.791±0.042→0.312±0.019,P<0.01). Microscopy showed that cells presented some morphologic characteristics of apoptosis.The flow cytometry indicated that after exposure to drugs,the apoptotic rates were higher in the co-treatment group with genistein and cisplatin.The effect in the co-treatment group showed a cell cycle blocking at G2/M phase.The immunocytochemical method showed that the expression levels of NF-κB were overexpressed after exposure to cisplatin (64.9±5.4)%, while the downregulation of NF-κB protein expression was seen in the co-treatment group[(23.4±2.1)%,P<0.01].CONCLUSION: Genistein combined with cisplatin can significantly suppress the growth of hepatocellular cancer cells through induction of apoptosis,which associates with the downregulation of NF-κB and may be useful for cancer chemotherapy.
