人脑星形细胞瘤组织中p57^(kip2)、p27^(kip1)的表达及与预后相关性

摘　　要：目的　研究p5 7kip2 、p2 7kip1在人脑星形细胞瘤发生、发展中的作用及与预后相关性。方法　采用免疫组织化学方法(S P法)检测40例星形细胞瘤及2 0例正常脑组织中p5 7kip2与p2 7kip1的表达情况。结果　40例星形细胞瘤中p5 7kip2蛋白、p2 7kip1蛋白阳性表达率分别为45 .0 % (18/ 40 )、5 2 .5 % (2 1/ 40 ) ,显著低于正常脑组织(75 .0 %、85 .0 % ) (P均<0 .0 5 ) ,两者表达均与星形细胞瘤恶性程度相关(P均<0 .0 5 )。p5 7kip2表达与p2 7kip1表达密切相关(γs =0 .5 0 4,P <0 .0 1)。p5 7kip2、p2 7kip1蛋白阳性表达患者的预后均优于阴性表达者(P均<0 .0 5 )。结论　p5 7kip2 、p2 7kip1蛋白表达在人脑星形细胞瘤发生起重要作用。两者与人脑星形细胞瘤患者预后的密切相关。Objective To explore roles of p57 kip2 and p27 kip1 in human brain astrocytoma and the prognostic significance.Methods The expression of p57 kip2 and p27 kip1 proteins in human brain astrocytoma tissues in 40 patients was detected by SP immunohistochemical technique.Results The p57 kip2 positive-expression rate in human brain astrocytoma was 45%,which was lower than that in normal brain tissue(P<0.05).The levels of p57 kip2 deletion between low malignant group(Ⅰ～Ⅱ) and high malignant group(Ⅲ～Ⅳ) were significantly different(P<0.05).The p27 kip1 positive-expression rate in human brain astrocytoma was 57.5%,which was lower than that in normal brain tissue(P<0.05).The p27 kip1 positive-expression correlated significantly with clinicopathologic grade(P<0.05).There were significant correlation between the expression of p57 kip2 and p27 kip1 in human brain astrocytoma(rs=0.504,P<0.01).Both p57 kip2 and p27 kip1 positive-expression correlated significantly with the prognosis of astrocytoma(P=0.0263,P=0.0123).Conclusion p57 kip2 and p27 kip1 protein plays an important role in the development of astrocytoma.They also correlated significantly with Prognosis.
