Cyclin D1和NF-κB在5-Fu诱导肝癌细胞凋亡过程中的表达

摘　　要：目的　探讨周期蛋白基因D1(CyclinD1)和核转录因子-κB(NF -κB)在5 氟脲嘧啶(5 - Fu)诱导肝癌细胞凋亡过程中的表达及其对细胞周期的影响。方法　体外培养人肝癌细胞株SMMC- 772 1,应用流式细胞术检测5 Fu对SMMC -772 1细胞周期和凋亡的影响;免疫印迹分析cyclinD1表达差异;用TransAM核转录因子活性检测试剂盒检测NF κB活性;细胞免疫组织化学检测NF κBp65入核情况。结果　5 Fu可诱导SMMC -772 1细胞凋亡,5 0、10 0、2 0 0 μg/ml 5 Fu分别作用2 4h后,其诱导细胞凋亡率分别为13 .2 % ,2 1.7%和3 5 .9% ,与对照组相比有显著性差异(P <0 .0 5 )。随着5- Fu浓度的增加,S期细胞比例和细胞增殖指数增加,CyclinD1蛋白的表达逐渐增强,NF -κBp65被激活并入核。结论　5 -Fu可诱导肝癌SMMC 772 1细胞凋亡;NF- κB在5 - Fu诱导肝癌细胞凋亡同时被激活;其下调基因CyclinD1过度表达可能是促进细胞增殖、对抗细胞凋亡的重要因素之一。Objective To explore the expression of cyclinD1,nuclear factor kappa B(NF-κB) and their effects on apoptosis induced by fluorouracil(5-Fu) in human liver cancer cell line.Methods The SMMC-7721 cells were cultured in vitro.Flow cytometry(FCM) was used to examine the effect of 5-Fu on the apoptosis and cell cycle of SMMC-7721 cell;cyclin D1 protein was tested by Western blot method,NF-κB activity of SMMC-7721 was measured by immunohistochemistry and an ELISA-based method.Results 5-Fu could induce apoptosis in a concentration-dependent manner.50?100?200 μg/ml 5-Fu resulted in an apoptotic rate of 13.2%?21.7% and 35.9%,respectively.As the concentration of 5-Fu increased,the percentage of S phase cells and proliferation index(PI) raised,and the cyclinD1 protein expression and NF-κB activity were also enhanced.Conclusion 5-Fu can induce apoptosis in SMMC-7721 and the NF-κB is actived simultaneously.Over-expression of cyclinD1 protein may be one important factor that promotes cell proliferation and plays a role in the process of anti-apoptosis.
