摘  要:目的 了解N 甲基D 天冬氨酸受体2A亚单位基因(NR2A)与精神分裂症的连锁关系。方法 选取NR2A亚单位基因所在区域的3个微卫星标志[D16s407、D16s3 075及NR2A启动子区的1个(GT)重复序列],对中国汉族81个独立精神分裂症受累同胞对及其家系成员共324个个体作基因分型,其中男166名,女158名,患病同胞对81个162例。采用受累同胞对法对分型资料进行非参数连锁分析。结果 324人的两点、多点非参数分析最大LOD值均位于D16s407,分别为2 643(P=0 004), 2 504(P=0 005),均大于2 2。结论 NR2A基因微卫星标志与精神分裂症存在提示性连锁关系,NR2A基因可能为精神分裂症的易感基因之一。Objective To investigate the linkage relationship between NR2A gene and schizophrenia.Methods Altogether 324 subjects from 81 families were recruited, including 162 patients and their relatives. The subjects were interviewed with the Diagnostic Interview for Genetic Studies and Family Interview for Genetic Studies. All enrolled patients fulfilled the Diagnostic and Statistical Manual of Mental Disorders, Fourth Edition criteria for schizophrenia. Three microsatellite polymorphisms from NR2A gene region were determined after the polymerase chain reaction. Nonparametric linkage analysis was performed with GENEHUNTER software version 2.1. Result Maximum nonparametric linkage score from tow-points and multi-points analysis were observed at marker D16s407,respectively was 2.643 (P=0.004),2.504 (P=0.005),which suggested NR2A gene for further studies. Conclusion Our data indicated that NR2A gene may be contribute to the genetic etiology of schizophrenia in Chinese population.
