摘 要:目的通过对于肝硬化动物肝脏葡萄糖激酶活性及mRNA的表达的观察,探讨其在肝硬化糖代谢异常中的作用。方法60只雄性SD大鼠,随机分为对照组和肝硬化组。应用6-磷酸葡萄糖法检测各组空腹及糖负荷后葡萄糖激酶活性,RT-PCR检测其mRAN表达。结果空腹时肝硬化大鼠肝脏葡萄糖激酶活性为0.70±0.391明显低于正常对照组的1.18±0.60。肝硬化大鼠肝脏葡萄糖激酶mRNA的表达为0.51±0.08,较正常对照组明显减少。糖负荷后,由于胰岛素的刺激,葡萄糖激酶活性为1.22±0.5,较空腹时明显增加,但与正常对照组的1.70±0.70仍显著下降。mRNA表达为0.58±0.11,较空腹时无明显变化。结论肝脏葡萄糖激酶mRNA表达的减少导致酶活性的下降可能为引起胰岛素抵抗从而导致糖代谢紊乱的原因之一。Objective To understand the mechanism of carbohydrate metabolism disturbance in liver cirrhosis rats by studying the change of the glucokinase. Methods 60 SD male rats were divided into two groups. The activity and the mRNA expression of glucokinase were measured. Results We found that the activity of the liver glucokinase and mRNA expression of liver glucokinase in the fasting situation was lower in cirrhosis group than that in the control group. The difference was significant. Although the activity of glucokinase was higher in both groups after taking glucose, it was significant lower in the cirrhosis group than in the control group. Conclusions The low glucokinase mRNA expression may results in the reduction of the activity of the glucokinase and play an important role in the glucose resistance.
