摘 要:采用多位点酶电泳(MEE)法对新疆近11年分离的154株伤寒沙门氏菌进行部分多位点醇基因型分类和群体遗传学研究.共测定分析7种代谢酶.研究结果表明,上述菌株共分为69个电泳型(ET),分属10个克隆系(CLa~CLj),其中80株爆发菌株分属21个ETs,3个CL(CLe、f和j),以ET17、20、21、24、26和27为主,占61.30%,CLf为优势克隆系,菌株占95.00%;74株散发菌株分属48个ETs,归为9个CL(CLa、b、c、d、f、g、h、i、j),没有发现菌株较为集中的ET,主要克隆CLh内菌株占47.30%,显示爆发和散发均可由多个ET型和克隆菌株引起,但爆发荫株的ET分布更为集中,有明显优势克隆.对上述菌株群体遗传结构分析表明:被测7种代谢酶基因位点均具有多态性,平均杂合度(h)为6.14,平均遗传多态值(H)为0.35.不同来源菌株相比h和H均无显著性差异(P均大于0.05).表明新疆不同来源的伤寒沙门氏菌可能来自同一群体.结果还显示每个酶基因位点都有一个优势等位基因,提示在伤寒沙门氏菌的进化演变过程中以自然选择为主。Multilocs enzyme electrophoresis( MEE)was employed to study the genotype taxonand population genetics of 154 isolatesof S. typhi from XinJiang in recent 11 years.Seven kinds of enzymes were examined anddetermined. One hundred and fifty-four strainsof S. typhi were classified into 69 electro- retic types(ETs ) and 10 clones(CLa-CLj). Among those 154 strains 80 epidemic isolatesbelonged to 21 ETs and 3 clones(CLe,f,j).ET17,20, 21 , 24,26, 27 were the main ETs(61.30%),and CLf was the dominant clone (95.00%).Seventy-four non-epidemic isolates belonged to 48 ETs and 9 clones(CLa,b,c,d,f,g,h,i,j).no obvious dominant ETwas found in them.and the main clone CLhaccounted for 43.30%. It demonstrated that bothoutbreak and sporadic epidemics could be causedby many strains of ETs and clones, butthe distribution of the former ETs was moreconcentrated and the preponderance of cloneswas more obvious.The analyses on structnreof popnlation genetics indicated thatall of 7 enzymes were polymorphic. Geneticdiversity among the enzyme loci was quite high.with an average of 0.35 per locus (H).Tht average mumber of alleles per locus(h)was 6.14.There was no significantdifference in h or H between the isolates ofdifferent sources(P> 0.05).It was clear thatS.typhi of different sources in Xinjiang wereoriginated from the same popnlation. Theresults also showed that there was a dominantallele in every enzyme locus.Natural selectionwas the main role in the evolution of S.typhi.
