野桑蚕12S rRNA基因的克隆及序列分析

摘　　要：通过PCR技术,对中国山东青州野桑蚕mtDNA的12SrRNA基因进行了克隆和序列分析。结果表明,青州野桑蚕12SrRNA基因为788nt,在DNA水平上,与家蚕12SrRNA基因的同源性达到99%,与中国陕西安康、日本筑波来源的野桑蚕12SrRNA基因的同源性分别为97%、98%;分子进化分析显示,家蚕可能是起源于中国野桑蚕。12S rRNA gene of mitochondrial DNA(mtDNA) from the Chinese Qingzhou Bombyx mandarina M. was cloned through PCR. The sequence analysis indicated that the 12S rRNA gene is 788nt in size,and shows 99% similarity with those of mtDNA from different species of B. mori,97% ,and 98% with that of Chinese Ankang and Japanese Tsukuba B. mandarina M on nucleotide level, respectively. Furthermore,the phylogenetic analysis illustrates that Bombyx mori possibly derived from Chinese Bombyx mandarina.
