摘 要:利用RT-PCR方法获得16个猪瘟流行毒株、5株弱毒疫苗株和外来的4株猪瘟E2基因部分编码序列的扩增片断,并对其进行测序,获得659bp含E2基因N段A、B、C、D四区域的编码序列。利用DNAstar软件对其中616bp的片段进行序列分析,并与Genebank中的Alfort﹑Brescia﹑HCLV﹑Shimen等毒株进行比较,结果20株病毒所测序列均为猪瘟E2基因序列,所有毒株碱基替换随机分布于整个序列,部分毒株出现碱基缺失现象。序列分析结果表明近期流行的毒株不仅与50年代流行的Shimen株、现用的疫苗株距离较远,而且流行毒株呈不同亚群方向演变。本研究所测的20株流行株E2基因N端6个与C端的2个半胱氨酸(Cys)均未发生变异,但其他位点的核甘酸变异与缺失,引起各区的抗原表位呈现不同程度的变异。The partial coding sequence of E2 gene of 20 hog cholera virus(HCV) field isolates and 5 attenu-ated vaccine strains were obtained by RT- PCR and sequenced. Including A、B、C and D domains, all sizewere 659bp.The obtained 616bp sequences were analysed by DNAstar and compared with sequences of Alfortstrain、Brescia strain、HCLV strain、Shimen strain and other reference strains in Genebank. The results showedthat those sequence fragments of 20 HCV field strains were the sequence of E2 gene of HCV compared withShimen strain,the base substitute of all strains were randomly distributed in the entire sequence and somestrains had base gap.The identity of nucleotide sequence and amino acid sequence of 13 HCV field strainswere 96.3%- 100%、96.6%- 100%,but compared with other 3 HCV field strains,the identity of neuclotide se-quence is only 85.9% - 89.3% .The identity of nuclotide sequence and amino acid sequence between theprovince and other provinces were 94.9%- 95.3%、95.9%- 96.9%.Between 5 HCV attenuated vaccine strains and20 HCV field strains,the homology of nuclotide and amino acid sequences were 81.8%- 82.6%、85.9%- 87.8%.Comparing 16 HCV field strains of the province with HCLV and Shimen strains,the identity of nucleotide se-quence were 81.1%- 83%、83.2%- 84%and that of amino acid sequences were 84%- 88.3%、86.9%- 92.2%.Those results indicated that HCV field strains prevalent in recent years in the province not only geneticallydivergent from the Shimen and vaccine strains,but also they evolved in different direction.It is showed epitopesbetween HCLV strains and field strains were variable in some extent by analysising the variation and gap ofsome amino acid residues,but the locations of 8 cysteire residues in obstained sequence of all were conserved.
