摘 要:目的观察人类端粒酶催化亚基(hTERT)反义寡核苷酸(ASODN)对Hela细胞增殖和细胞周期的影响。方法采用四甲基偶氮唑蓝(MTT)法检测ASODN对Hela细胞增值能力及细胞生长的影响,用流式细胞仪检测细胞周期和细胞增殖指数。结果不同浓度的ASODN转染细胞后,均不同程度地影响细胞的生长。ASODN作用细胞后,使细胞周期发生变化,表现为G0/G1期细胞增多,S期细胞减少,细胞增殖指数下降。结论以hTERT基因为靶点的反义寡核苷酸能抑制Hela细胞的体外生长,使细胞阻滞于G0/G1期。Object To investigate the effects of human telomerase reverse transcriptase(hTERT) antisense phosphorothioate oligodeoxynucleotide (ASODN) on cell proliferation and cell cycle kinetics of Hela cell line in vitro. Methods We examined the effects of the growth of cell by MTT assay. Cell cycles were detected by flow cytometry(FCM) analysis technology. Results After treatment with ASODN of hTERT with different doses of 0.05 μmol/L, 0.1 μmol/L and 0.2 μmol/L respectively for 72 hours, the inhibitory growth rate of Hela cell was 6.74%, 13.12% and 22.01% respectively. ASODN of hTERT could change the cell cycle of Hela cell line, G0/G1 phase cells increased, S phase cells decreased and the proliferation index of Hela cell decreased. Conclusion In vitro, ASODN of hTERT possess inhibitory effects of proliferation and growth in Hela cell line and arrest the cell cycle progression in G0/G1 phase.
