摘 要:以一株能转化鹅去氧胆酸(chenodeoxycholicacid,CDCA)为熊去氧胆酸(ursodeoxycholicacid,UDCA)的厌氧菌-ClostridiumperfringensTypeAHS-10(产气英膜梭菌HS-10)作为试验菌,进行细胞固定化,研究不同的吸附和包埋材料制备的固定化细胞对UDCA生物转化的影响,探索提高底物浓度、菌种底物耐受度及转化率的可能性。实验结果表明,海藻酸钙、多孔陶瓷和蛭石都是较为理想的固定化材料,特别是先将菌体在含1.2mmol/L的RCM(reinforcedclostridiummedium)培养基中进行"预培养",然后再经2%的海藻酸钙固定化后能显著提高底物耐受度,当底物浓底高达1.0mmol/L和2.0mmol/L时,最大转化率分别为40%和20%Clostridium perfringens HS-10 was immobilized to study the biotransformation ofchenodeoxycholic acid(CDCA) to ursodeoxycholic acid in order to increase the substrateconcentration ,the anaerobe tolarence to substrate and the bioconversion rate, the iniinobilzedcells were made with ditferent adsorbing and embedding materials. It was found tbat calciumalginate, porous eramic and vermiculite were perfect immobilizing materials if the anaerobe HS -10 was 'pre-cultured'in RCM medium containing 1.2mmol/L CDCA, the tolerence tosubstrate could be remarkably mised when the bacterial cells were embedded with 2% calciumalginate.The greatest conversion rates were 40% and 20% with substrate concentiations up to1.0mmol/L and 2.0 mmol/L.
