含水稻rbcL重组质粒限制图谱的构建

摘　　要：从水稻叶绿体基因文库中挑选合rbcL基因的3号克隆,采用大量煮沸法,分离纯化了质粒DNA,利用限制性核酸内切酶BamH I、 Sma I、Xhol和Cta I进行单酶或双酶切,在BamH I限制图谱的基础上构建了几种限制酶的限制图谱.Pcos 2-γ-3 containing the gene for the rbcL is selected from the cosmid library of rice chloroplast. Pcos 2-γ-3 plasmid DNA is isolated and purified by boiling large scale lysis method. Restriction map of the recombinant plasmid is constructed by eleavage with several restriction endonucleases, such as BamH I 、Sma I、Xho I、 depending on the restridtion map of BamH I.
