摘 要:目的探讨β胡萝卜素(β-Carton,β-C)与维生素E(VitaminE,VE)不同配比方案,对荷瘤小鼠放、化疗后血液学及肿瘤生长的影响,确定其减轻放、化疗毒副作用的合理剂量配比水平。方法采用有重复的正交实验设计,用L8(27)正交表研究β-C、VE不同水平的两两配比及放疗、化疗2种治疗方案的不同配比对荷瘤小鼠放、化疗后血液白细胞、血小板及瘤重的影响。24只小鼠随机分为8组,经口给予不同剂量配比的β-C、VE,20天后每只小鼠腋下接种S-180瘤液0.2ml,接种瘤液后第2天对小鼠分别进行化疗、放疗。放疗组为一次性全身γ-射线辐射,剂量为4Gy,化疗组给予环磷酰胺,剂量为180mg/kg·bw,隔日1次,共5次。治疗前一天及治疗后第3天进行白细胞计数及血小板计数,治疗后第10天解剖动物,检测动物的肿瘤重量,计算瘤体比。结果单因素β-C在抑制由于放、化疗引起的白细胞、血小板下降及抑制肿瘤生长方面作用明显(P<0.05);单因素VE可明显抑制荷瘤小鼠的肿瘤生长(P<0.05);β-C和VE交互作用可抑制荷瘤小鼠由于放、化疗引起血小板下降(P<0.05),并明显抑制荷瘤小鼠肿瘤生长(P<0.05)。结论β-C+VE在抑制荷瘤小鼠由于放、化疗引起血小板下降方面合理的剂量配比为β-C60mg/kg+VE50mg/kg;β-C+VE抑制荷瘤小鼠肿瘤生长的合理剂量配比为β-C30mg/kg+VE50mg/kg(以每公斤体重计)。Objective To get the best combination plan of β-Carotene and vitamin E. Different combination plans' effects on haematology and the tumor weight of mice treated by chemotherapy or radiotherapy was explored. Methods Othogonal design (L8(2^7)) was used to observe the effects of the different combination plans of β-Carotene and vitamin E on haematology and the tumor weight of mouse after chemotherapy or radiotherapy. First, 24 mice were divided into 8 groups randomly and oral administered by different dose combination of β-Carotene and vitamin E. On the 21th days, the tumor cell (S-180) was implanted into the subaxillary tissue of mice. In the following day they were treated by chemotherapy (180 mg/kg·Bw/d×5 times), once every two days) or radiotherapy (4 Gy×1). The leucocytes and platelets were counted one day before treatment and at the 3 rd day after treatment. At the l0 th day after treatment, mice were sacrificed, and tumor was weighted. Results β-Carotene could restrain the decrease of the leucocyte and the growth of tumor (P〈0.05). VE could inhibit the growth of tumor obviously too (P〈0.05). The combination of β-C and VE restrained the decrease of platelets and inhibit the growth of tumor evidently(P〈0.05). Conclusion The combination plan of β-Carotene (60 mg/kg·bw) and VE (50 mg/kg·bw) was better on restraining the decrease of platelets, and 30 mg/kg·bw of β-C with 50 mg/kg· bw of VE, was better on inhibiting the growth of tumor.
