人结肠癌耐药细胞系HCT_(V2000)的建立

摘　　要：HCT－8是一种体外培养的人结肠癌细胞系。本文用长春新碱选择得到对多种抗肿瘤药物交叉耐药的HCTv2000细胞系。与HCT－8细胞相比，HCTv2000对长春新碱的耐药程度为147倍，秋水仙碱为65倍，高三尖杉酯碱为47倍．紫杉醇为31倍，阿霉素为20倍，对鬼臼乙叉甙等也产生了耐药性，但对5-氟尿嘧啶的敏感性无明显改变，符合多向耐药表型特点。HCTv2000细胞生长状态良好，耐药性能稳定，可以用作分析耐药机制的模型。HCT-8 is a cultured cell line of human colon cancer. In the present study, HCTv2000 line was isohited from the cells by vincristine selection, showing cross- resistance to various anticancer drugs. HCTv2000 cells exhibited 147-fold more resistance t0 vincristine, 65-fold to colchicine, 47-fold to homoharringtonine, 31-fold to taxol, and 20-fold to adriamycin,as compared with HCT8 cells. This line of cells also showed resistance to VP-16, but the sensitivity to 5 - Fu appeared to be not changed significantly. This phenotype is in agreement with the multi-drug resistance. HCTv2000 cells are growing well in vitro with stable drug-resistant characteristics and may be useful as a model for analysing mechanisms of drug resistance.
