摘　　要：目的探讨组蛋白去乙酰化酶抑制剂LBH589对胃癌细胞系BGC-823增殖的影响。方法体外培养胃癌BGC-823细胞,MTT检测LBH589对BGC-823细胞增殖率的影响;Western Blotting检测内质网应激相关蛋白GRP78,内质网凋亡相关蛋白CHOP、caspase-4表达水平的改变。结果与对照组相比,给予不同浓度LBH589细胞增殖率明显下降;同时LBH589可以引起内质网应激相关蛋白GRP78表达水平的上调,内质网凋亡相关蛋白CHOP、caspase-4表达水平也明显升高。结论组蛋白去乙酰化酶抑制剂LBH589可能通过内质网应激-凋亡通路诱导BGC-823细胞发生凋亡。Objective To investigate the effect of histone deacetyltransferase inhibitor LBH589 on cell proliferation of stomach cancer BGC-823 cells.Methods MTT assay was employed to examine the growth suppression of the cells.Western Blotting was used to detect the expressions of Grp78,CHOP and Caspase-4.Results MTT assay showed that vary dosages of LBH589 decreased the growth rate of BGC-823 cells.LBH589 could increase the expressions of Grp78,CHOP and Caspase-4.Conclusion Histone deacetyltransferase inhibitor LBH589 could induce the apoptosis of stomach cancer BGC-823 cells through endoplasmic reticulum stress(ER stress)pathway and decrease the proliferation rate.
