摘  要:目的建立体外培养和扩增树突状细胞(dendritic cell-DC)的方法,并进行形态学观察. 方法在无菌条件下提取BALB/c小鼠骨髓细胞,分离单个核细胞,以小鼠重组粒细胞-巨噬细胞集落刺激因子(GM-CSF)和白细胞介素(IL-4)协同诱导下培养,免疫组织化学染色、光镜和电镜下观察树突状细胞的形态. 结果体外培养2周后小鼠树突状细胞可达70%~80%,光镜下可见典型的树突状细胞形态,S-100蛋白染色均呈阳性.电镜下,培养的细胞具有典型树突状细胞的形态特征,细胞功能活跃. 结论体外大量培养和扩增小鼠骨髓树突状细胞,具有典型的树突状细胞的形态,为研究树突状细胞的功能和表型以及抗肿瘤作用提供体外实验材料.Objective To establish a method of cultivation and purification of dendritic cells (DCs) from mouse bone marrow in vitro and observe their morphology. Methods Mouse bone marrow cell suspensions were prepared with aseptic technique, then uninuclear cells were isolated and induced in vitro by both granulocyte-macrophage colony-stimulating factor(GM-CSF) and interleukin-4( IL-4), and their morphology was observed by light, electron microscopy and immunohistochemistry. Results The percentage of the cultured DCs was 70% - 80% with typical morphology of DCs under light microscope and S-100 protein positive staining 2 weeks after culture. Under electron microscopy, these cells appeared a typical morphological feature of DCs and active function. Conclusion A large number of DCs from mouse bone marrow in vitro exhibited typical morphology of DC, which might be an experimental material for further study on function, marker and anti - tumor role of DCs in vitro.
