摘  要:本研究以各种不同性质的氨基酸为配基通过共价键将其固定在纤维素凝胶微球上制备了类风湿关节炎吸附剂,研究了间隔臂长度和种类对吸附剂吸附性能的影响。结果表明以苯丙氨酸(Phe)或色氨酸(Trp)为配基的吸附剂对类风湿因子有较高的清除率,是较好的配基;吸附剂的间隔臂对其吸附性能有较大影响,随着间隔臂的加长,吸附剂对类风湿因子的清除率相应增加:以Phe为配基时对IgGRF的清除率由48.7%增加到59.6%,以Trp为配基时对IgGRF的清除率由46.2%增加到60.1%:采用支化间隔臂可明显提高配基的固定量,吸附剂对类风湿因子的清除率亦显著提高(以Phe为配基时对IgGRF的清除率为74%)。以长耳白兔为动物模型进行体外血液灌流实验显示此类吸附剂对血液中血细胞、血浆蛋白、血浆电解质的影响很小,该吸附剂的血液相容性良好。Rheumatoid arthritis adsorbents were prepared by immobilizing various kinds of amino acids to cellulose gel beacls. The effects of spacers on adsorption properties were investigated in this article. The results showed that the adsorbents with phenylalanine or tryptophane ligand obtained high adsorption percentage for rheumatoid factors(RFs) . The adsorption capacities were significantly increased with the spacers. Adsorbents with the dendrimer spacer obtained higher immobilized amount of ligands and higher adsorption percentage for rheumatoid factors(74% with phenylalanine ligand) compared to those with other kinds of spacers. Hemoperfusion experiments were carried out in vitro with rabbits. It was shown that the adsorbents had little negative effects on blood cells, plasma albumin and electrolyte, the adsorbents exhibited good blood compatibility.
