摘 要:Fez1基因是由IshiiH发现的一个新的候选抑癌基因。它定位于染色体8p22,编码67kDa的亮氨酸拉链蛋白,有与依赖cAMP激活蛋白相似区域。它在人类的许多肿瘤包括膀胱移行细胞癌,前列腺癌,食管癌,乳腺癌,胃癌等都存在异常表达。但原因尚不清楚,可能包括有:①基因缺失;②无义突变或点突变;③反常甲基化;④等位基因丢失所致的反常转录。这些改变可能在肿瘤的发生发展中起重要作用。近年来通过对其功能的研究,认为Fez1基因的功能有:抑制肿瘤增殖,调节细胞周期,与微管调节相关。虽然对Fez1基因的研究取得一些成绩,但仍需研究其功能,失活的原因,以及在肿瘤中发生发展的作用和机制。Fezl is a new candidate tumor suppressor gene discovered by Ishii H. It located on chromosome 8p22 and encodes a 67 - kd leucine - zipper protein with a region of similarity to cAMP - dependent activated protein. Fezl alterations,which include absent or reduced, have been reported in many human malignancies, including bladder transitional cell carcinoma, prostate, esophageal, breast, and gastric carcinomas. But the mechanism of alterations of Fezl expression in human cancers has not been demonstrated convincingly: maybe including:①genomic deletions; ②missense mutation or point mutation ;③aberrant methylation ;④aberrant transcripts of the allelic losses. These change may play a role in the development of tumor ,The function of Fezl gene include :inhibit tumor growth , regulate cell cycle, associate with the regulation of microtubule. Although there have got some achievement, more effort should be required to study the function of the Fezl gene , reason of it's silence , the role and mechanisms in the development of tumor.
