摘 要:根据人和大鼠的腺苷一磷酸脱氨酶1(AMPD1)基因序列设计引物,采用PCR-RF-SSCP技术检测了130只北京油鸡AMPD1基因多态性,并分析了AMPD1的不同基因型与北京油鸡肌苷酸含量的关系。结果表明:在北京油鸡中检测到AMPD1基因的A、B、C3个等位基因,基因频率分别为0.180 8、0.661 5、0.157 7;产生AA、AB、AC、BB、BC、CC6种基因型,基因型频率分别为0.076 9、0.176 90、.030 80、.461 5、0.223 1、0.030 8。最小二乘分析表明基因型AC、BB所对应的肌苷酸含量最小二乘均值显著高于基因型AA所对应的最小二乘均值(P<0.05),肌苷酸含量在AMPD1其余基因型之间没有显著差异(P>0.05)。Adenosine monophosphate deaminase 1 (AMPD1) gene was studied as a candidate gene of inosine monophosphate (IMP) content of Beijing fatty chickens. According to the sequence of AMPD1 gene of human and rat, primers were designed to amplify the AMPD1 gene of chicken. Based on the clone sequencing, single nucleotide polymorphism of AMPD1 gene was detected in 130 Beijing fatty chickens by PCR-RF-SSCP. The results showed that three alleles (A,B,C) and six genotypes (AA,AB,AC,BB,BC, CC) were found in Beijing fatty chickens. Frequencies of alleles A, B, C were 0. 180 8, 0. 661 5, 0. 157 7, respectively. Frequencies of genotypes AA, AB, AC, BB, BC, CC were 0. 076 9, 0. 176 9, 0. 030 8, 0. 461 5, 0. 223 1, 0. 030 8,respectively. The least squares means of IMP content for AC and BB were higher than that for AA (P〈0.05), and there were no significant differences between other genotypes (P〉0. 05).
