摘 要:目的:探讨HBV转录调控核心序列在肝细胞中转录的活性。方法:构建HBV增强子和启动子调控的荧光素酶报告载体,用脂质体转染法将其转染肝细胞和非肝细胞,双荧光素酶报告基因试验检测其在不同细胞中的转录活性。结果:HBV启动子在2.2.15细胞中转录活性最高,在其他肝细胞中的转录活性显著高于非肝细胞。结论:由HBV启动子构建的载体可能是一种新颖的有前景的靶向性肝癌细胞基因治疗载体。Objective:To study the hepatic-specific effect of HBV transcriptional regulatory sequence. Methods: Luciferase expression vector ( pEN Ⅱ WLuc) driven by HBV promoter was constructed. The recombinant plasmids were transfected into five hepatic cells and five non-hepatic cells using LipofectamineTM reagent. The luciferase expression which indirectly represented the transcriptional activity of HBV promoter lying upstream of Luc gene was detected with Dual-Luciferase Reporter Assay System. Results :The results showed that the HBV promoter had high transcriptional activity in hepatic cells, particularly in 2.2.15 cells, but had low transcriptional activity in non-hepatic cells. Conclusion:The hepatic-specific expression vector affording HBV promoter may be a novel and promising targeting approach for the treatment of hepatic carcinoma.
