摘 要:以介孔分子筛MCM48为载体、戊二醛作交联剂对木瓜蛋白酶进行了固定化。实验结果表明,与游离酶相比,固定化酶的热稳定性有了显著的提高,其半衰期达到了2500min以上,是游离酶(80min)的30倍以上。固定化酶的pH稳定性和储藏稳定性也有了明显改善。此外,固定化酶还具有很好的操作稳定性,经过12d的连续酶解,酶活力仍保持在初始酶活力的70%以上。The present work dealt with the immobilization of papain on the mesoporous molecular sieve MCM- 48 with the aid of a coupling agent, glutaraldehyde. The thermostability of immobilized enzyme was greatly improved. The half-life of the immobilized enzyme was more than 2 500 min, which was significantly higher than that of free enzyme (about 80 min). The influence of pH on immobilized enzyme activity was less than on free enzyme. The immobilized enzyme also showed preferable storage stability as compared with free enzyme. Moreover, the immobilized enzyme exhibited good operational stability, its activity was still over 70 % based on the initial activity after 12-day continuous operation.
