摘 要:目的:研究绿茶提取物GTC对致癌剂DMBA诱发的SD大鼠乳腺癌前增生的抑制作用并初步探讨其可能的作用机制。方法:170只7周龄雌性SD大鼠,按体质量分层后随机分为4组,即A(空白对照组)、B(模型组)、C(GTC对照组)和D(实验组)。于实验开始日,B组和D组每只大鼠接受一次性DMBA油溶液灌胃,剂量为7mg/100g体质量;同时C组和D组每只大鼠接受GTC水溶液灌胃,剂量为每只80mg/d,A组和B组大鼠自由饮用自来水。于实验第4、8、12、16和20周末分批处死各组大鼠,取乳腺组织常规制片,HE染色;常规SP免疫组化染色和TUNEL检测20周末大鼠乳腺导管上皮细胞增殖指数和凋亡指数。结果:饮茶组(C组和D组)和非饮茶组(A组和B组)相比,其增生性病变较少;B组和D组之间差异有统计学意义,P值分别为0.024、0.001、0.000、0.001和0.000,而在对照A组和C组之间差异无统计学意义,P值分别为0.947、0.263、0.879、0.133和0.274;B组和D组之间增生性病变计数的差值呈现随时间延长而增加的趋势;20周末,D组大鼠乳腺导管上皮细胞增殖指数(0.19±0.12)显著低于B组(0.48±0.20),t=4.106,P=0.001;D组大鼠乳腺导管上皮细胞凋亡指数(0.57±3.74×10-2)稍高于B组(0.53±0.10),但差异无统计学意义,t=-1.396,P=0.177。结论:GTC对DMBA诱导的大鼠乳腺导管上皮细胞癌前增生具有一定的抑制作用且这种抑制作用呈现时间依赖性递增;GTC抑制大鼠乳腺导管上皮细胞癌前增生主要通过抑制细胞的增殖活性而非通过诱导细胞凋亡的途径。OBJECTIVE: To illustrate the inhibitory effect of GTC (green tea eateehins) on the development of the hyperplastie lesions in rat mammary gland induced by DMBA and the possible mechanism. METHODS: A total of 170 7-week old female SD rats was divided into4 groups randomly: group A (blank control), B (DMBA gavage). C (GTC garage) and D (DMBA/GTC gavage). At the beginning of the experiment. Group B and D received a 7 mg ·100 g bodyweight intragastrie dose of DMBA, at the same time. group C and D received one intragastric dose of GTC (80 mg rat day). The GTC-gavage was administrated. All rats got the basic fodder and tap water freely. By the end of the 4th, 8th, 12th, 16th, and 20th, all the rats in every group were sdlected randomly sacrificed averagely for each time point, the breast tissues were removed and fixed and made HE sections rouinely. Expressiou of PCNA was detected bv S-P immunohistochemical staining and the apoptosis was ,letected by TUNEL technique for the rats in group D and B of the 20th week. RESULTS: There were more hyperplastic lesions which include solid bodies, buds. papil lary hyperplasia and mitosis in group B and D than those lesions in group A and C. Such difference between group Band Dis statisti cally significant (P〈0.05). hut no significance can be found be tween group A and C. The difference between D and Bin the hyperplastic lesions is increasing with the time passing. The proliferation index in group D is significantly lower than that in group B (0. 19%±0. 12 and 0.48±0.20 respectively) ,P=0.001 ; but no sta tistical significance is found for the apoptosis index between group Band D (0.53±0. 10 and 0.57±3.74×10^2 respectively), P= 0. 177. CONCLUSIONS: From this study, we can suppose that constant GTC-gavage can inhibit the development of the breast ductal hyperplasia induced by DMBA in SD rat to some extent and such inhibitory effect seems enhancing with the time passing; The mechanism of GTC inhibitory effect seems to be the inhib
