脑胶质瘤增殖细胞核抗原和Cyclin Dl蛋白的表达及生物学意义

摘  要:目的探讨增殖细胞核抗原(PCNA)和CyclinDl蛋白在脑胶质瘤中的表达及其在胶质瘤发生、发展过程中的生物学意义。方法对已明确诊断的82例脑胶质瘤及10例瘤旁正常脑组织利用S-P免疫组化法检测PCNA和CyclinDl蛋白。结果82例胶质瘤中PCNA标记指数(PCNALI)较正常脑组织明显升高,并随胶质瘤恶性程度的升高而呈上升趋势(P<0.05)。CyclinDl蛋白阳性率在正常脑组织及肿瘤组织间以及不同分级的胶质瘤间无明显差异(P>0.1)。结论PCNA是胶质瘤发生和恶性转化的重要促进因子,而CyclinDl可能不参与该类肿瘤的发生和发展。Objective To explore the abnormalities of expression of PCNA,Cyclin D1 protein and their biological significance in human gliomas.genesis and development. Methods 82 cases of glioma samples and 10cases of normal brain tissue samples adjacent to the tumour were detected by S-P immunohistochemical staining. Results PCNA Labeling index are higher than those in normal brains and increased with the grade of human gliomas. There was neither significant difference in the expression of Cyclin D1 between normal brain tissue and glioma samples nor among different grades. Conclusion The abnormalities of the expression of PCNA gene are the important factors for the development of glioma and can be used as indexes for judgement about pathological grade in brain glioma;But the expression of Cyclin D1 protein probably shows no relationship with the glioma dovelopment and progression.
