摘 要:目的研究音乐刺激对海马神经元GABA_A受体表达的影响。方法在大鼠生长发育的不同时期,给予音乐刺激,用免疫组织化学和PCR技术及图像分析系统定量研究音乐刺激后,大鼠海马神经元GABA_A受体及其编码该受体的mRNA的表达。结果音乐刺激后,大鼠海马神经元GABA_A受体免疫染色的光密度比对照组增加,在CA1区分别从对照组(32.21±4.23)增加到持续音乐组(44.72±6.21),生后音乐组(43.61±6.12),胎期音乐组(34.34±4.54);编码受体的mRNA表达相对量增加,从对照组(0.67±0.23)增加到持续音乐组(0.99±0.34),生后音乐组(0.92±0.32),胎期音乐组(0.73±0.25),这些变化以持续音乐刺激组最明显,生后音乐刺激组次之,胎期音乐刺激组和对照组最弱。结论音乐刺激能增强大鼠海马神经元GABA_A受体及编码该受体的mRNA表达,其增强作用具有刺激时间依赖性关系。Objective To study the effect of music on the expression of GABAA receptor and the mRNA coding the receptor on neurons from rat hippocampus. Methods The rats were exposed to the music in their different developing stages, and the immunohistochemistry and PCR technique were used to demonstrate the expression of GABAk receptor and its mRNA on hippocampal neurons from rat quantitatively with the aid of image analysis system. Results After music exposure the optic density of GABAg receptor on hippoeampal neuron in area CA1 increased from 32.21 ± 4.23 in control group to 44.72 ± 6.21,43.61 ±6.12,34.34 ±4.54 in persistent, postnatal and prenatal groups respectively, and the mRNA coding GABAA receptor upregulated from 0.67 ± 0.23 in control to 0.99 ±0.34,0.92 ±0.32 and 0.73 ±0.25 in persistent, postnatal and prenatal groups respectively. Expression of both GABAA receptor and its mRNA changed the greatest in the persistent music exposure group, then in the postnatal and the least in prenatal and control groups. Conclusion The music can improve the expression of GABAA receptor and its mRNA on hippoeampal neurons from rat in a stimulating time-dependent manner.
