高强度超声对宫颈癌小鼠再次接种同种肿瘤的抑制作用

摘　　要：目的探讨高强度超声(HIU)治疗对宫颈癌小鼠再次接种同种肿瘤的抑制作用。方法72只接种成功的U14宫颈癌小鼠随机分为HIU组、手术组和对照组,每组24只,于接种肿瘤后第7天分别接受HIU治疗、手术治疗及HIU假治疗;于接种肿瘤后第17天,每组小鼠再随机分为3组,分别再次接种2×106(n=8),2×107(n=8)或0(n=8)个U14肿瘤细胞,记录各组小鼠的无癌存活数。结果HIU组、手术组和对照组再次接种2×106个U14肿瘤细胞,其无癌存活率分别为100.0%,75.0%和0.0%,3组比较差异有统计学意义(P<0.05);接种2×107个U14肿瘤细胞时,无癌存活率分别为87.5%,25.0%和0.0%,3组比较差异有统计学意义(P<0.05);未行第2次肿瘤接种的小鼠无癌存活率,HIU组(100%)、手术组(100%)均较对照组(0%)差异有统计学意义(P<0.05)。结论HIU治疗小鼠宫颈癌是有效的,HIU较手术治愈的小鼠对同种肿瘤有更强的抑癌作用,这可能是由于HIU杀死的肿瘤细胞起到了“HIU原位固化瘤疫苗”作用。Objective To investigate the inhibitive effects of high intensity ultrasound （HIU） on the second inoculation of the same strain tumor cells in mice with U14 cervical cancer. Methods Seventy-two mice with U14 cervical cancer were divided into three groups ： a HIU group （ group A, n = 24）, an operation group （ group B, n = 24） and a sham-HIU group （ group C, n = 24） , and treated with HIU （50 Watt/cm^2 ） , operation and HIU （0 Watt/ cm^2） 7 d after inoculation, respectively. The mice in each group were divided into three sub-groups（ n = 8） and reinoculated with 2 ×10^ , 2 ×10^7 or 0 U14 cancer cells at 17 d after first inoculation, and then the rate of survival mice without tumor （RSWT） in every group was recorded at 150 d after second inoculation. Results RSWTs in group A, group B and group C were 100.0% ,75.0% and 0.0% after re-inoculation of 2 ×10^6 U14 cancer cell, 87.5% ,25.0% and 0.0% after re-inoculation of 2 ×10^7 cells, and 100% , 100% and 0% without re-inoculation, respectively. Conclusion It was effective for HIU to treat cervical cancer in mice, furthermore the effects of HIU, playing a role as ＂ HIU solidified tumor vaccine＂ , on the second inoculation in mice with U14 cervical cancer might be more valuable than those of operation.
