摘 要:目的:研究检测caspase-3、多聚ADP-核糖聚合酶(PARP)蛋白在人肺癌组织的表达及与临床病理特征的关系。方法:在57例人肺癌原发灶中应用第二代免疫组织化学E liv ision法检测caspase-3、PARP蛋白的表达。结果:在57例肺癌原发灶中caspase-3、PARP两种蛋白表达阳性率分别为66.67%、33.33%。caspase-3与细胞分化程度(P=0.023)、淋巴结转移(P=0.004)和组织学类型显著相关(P=0.036),PARP与淋巴结转移显著相关(P=0.025)。57例人肺癌组织中,caspase-3的表达与PARP呈显著负相关(P=0.001)。结论:caspase-3通过降低其底物PARP的表达从而促进肿瘤细胞的凋亡明显地抑制了肺癌的浸润和转移。Objective: The current study was designed to investigate the relationship between the expressions of caspase-3, PARP proteins and the clinicalpathological characteristic of pulmonary carcinoma. Methods: The serial sections of the same tissues were also subjected to immunohistochemical staining of caspase-3 and PARP in 57 patients with pulmonary carcinoma (21 patients with squamous cell carcinoma, 27 with adenocarcinoma, 4 with large cell carcinoma and 5 with small cell lung carcinoma) . Results: In 57 pulmonary carcinomas, caspase-3 and PARP proteins positive rates were 66. 67%, 33. 33% respectively. The positive expression of caspase-3 protein was highly associated with cellular differentiation degree (P = 0.023), lymph node metastasis(P=0.004) and the pathological types(P=0.036). DFF45 expression was only associated with lymph node metastasis(P= 0. 025). In 57 pulmonary carcinomas, a negative correlation were present between caspase-3 and PARP protein expression(P=0. 001). Conclusion:Caspase-3 obviously inhibits invasion and metastasis of pulmonary carcinoma by extensively degrading target proteins such as PARP.
