摘  要:背景与目的:评价三羟异黄酮对黑色素瘤B16细胞的诱导分化作用。材料与方法:以4’,5,7三羟异黄酮处理黑色素瘤B16细胞1~4d后,以生长曲线反映其增殖活力,以B16细胞形态、黑色素含量、及流式细胞仪检测细胞周期变化等观察三羟异黄酮对黑色素瘤B16细胞的诱导分化作用。结果:用10、30、90μmol/L的三羟异黄酮作用肿瘤细胞,有不同程度的抑瘤作用。表现为黑色素生成能力增加,细胞生长缓慢。可使B16细胞阻断在S期。结论:三羟异黄酮不同剂量(10、30、90μmol/L)对B16细胞均有明显的分化诱导作用,对体外黑色素瘤增殖有明显的抑制作用。BACKGROUND & AIM: To evaluate the inhibiting effects of Genistein on Bl6 melanoma cells. MATERIAL AND METHODS: Bl6 melanoma cells were treated with 10, 30, 90μmol/L of Genistein for 1- 4 days. Growth curve test served as the index to reflect proliferative activity of B16 melanoma cells .Morphological changes of B l6 melanoma cells, melanin content in the supernatant of melanoma cells, distribution of cell cycle were assessed by Flow Cytometery. RESULTS: Genistein at the concentration of 10,30,90 μmol/L inhibited significantly the growth of B16 melanoma cells in a close dependent manner with statistical significance (P 〈 0.05 vs control). Interestingly, Genistein induced the differentiation of B16 melanoma cells and decreased the melanin granule and number of cells. Flow cytometry showed that numerous cells were arrested at S phase of cell cycle. CONCLUSION: Genistein at the dose of 10, 30, 90μmol/L significantly induced differentiation of B16 cells while inhibiting their growth.
