ras癌基因点突变与膀胱癌关系的研究

摘　　要：采用聚合酶链反应（ＰＣＲ），结合限制性片段长度多态性（ＲＦＬＰ）分析的方法，检测２４例石蜡包埋的膀胱癌组织中１５例Ｈａ－ｒａｓ癌基因第１２位密码子的点突变呈阳性（６２．５％），提示Ｈａ－ｒａｓ癌基因第１２位密码子的点突变随膀胱癌的级别、期别升高，可作为判断肿瘤恶性程度、进展和预后的监测指标。Point mutation at codon 12 of Ha-ras gene was assayed by polymerase chain reaction together with assay of restriction fragment lenth polymorphism(RFLP)was carried out in 24 paraffin-embeded specimens of human bladder cancer,The positive mutation rate was 62. 5%(15/24). The mutation rate increased with the rise of tumor grade and stage and might serve as a marker for the malignancy and prognosis of bladder cancer.
