摘  要:目的 建立大鼠腹主动脉、下腔静脉暂时性阻断、半身隔离灌注的实验模型,并探讨该模型的安全性。方法选体质量250-300g成年SD大鼠24只,随机分成A、B、C、D组,采用暂时性阻断腹主动脉、下腔静脉血流,在其阻断的下方分别插管,连接一恒流泵,作引流、灌注,建立后半身隔离性循环灌注模型。A、B、C、D组分别阻断灌注30、60、90、120min,观察阻断隔离灌注前、中、后的血压、呼吸、心率的改变;灌注前后血Na^+、K^+、Ca^2+、Cl^-及RBC、WBC计数的变化及大鼠的恢复情况。结果阻断腹主动脉、下腔静脉时大鼠颈动脉血压出现短暂小幅度上升,数分钟后恢复正常并维持到隔离灌注结束,阻断隔离灌注前后呼吸、心率。大鼠血Na^+、K^+、Ca^2+、Cl^-及RBC、WBC计数均无明显变化。A、B组大鼠精神、饮食、运动恢复情况良好;C组5只大鼠灌注术后出现双侧后肢不全瘫,2周后大部分恢复;D组均出现双侧后肢瘫痪,2周后部分恢复。结论利用大鼠可以建立简单、廉价的腹主动脉、下腔静脉阻断半身隔离性循环灌注动物模型;腹主动脉、下腔静脉阻断半身隔离性循环灌注持续60min是安全的。是进行区域性阻断灌注治疗研究的理想动物模型。Objective To establish the experimental rat model by temporarily blocking the distal abdominal aorta and inferior vein cava, and isolated perfusion circulation. Methods Twenty-four SD rats, weigthing 250-300 g, were randomly divided into group A, B, C, and D. The blood flow was blocked by clamping abdominal aorta and vein, then a tube was inserted into the isolated part of posterior semibody by conneting tubes and a pump. Isolated perfusion time of group A,B,C,D were 30,60,90 and 120 minutes respectively. The blood pressure, heart rate, respiratory rate, ion in plasma, RBC and WBC count in plasma, were measured berfore, during, after isolated perfusion,and the recovery course of rat health was recorded. Results Temporary increase of blood pressure oct .uFred after blocking the blood flow of abdominal aorta and vein. The respiratory rate,heart rate,ion in plasma,RBC and WBC count in plasma didn't change obviously before and during isolated perfusion. The spirit, drink, eating and posterior extremity movement of group A, B recoveriedquickiy; Incomplete paralysis of posterior extremity occurred in 5 rats of group C, but recovered to normal in 2 weeks after isolated perfusion;Complete paralysis of posterior extremities occurred in group D and recovered partialy in 2 weeks after isolated perfusion. Conclusions Experimental rat model of isolated perfusion of posterior semibody by blocking abdominal aorta and vein can be established. It will be safe to block distal abdominal aorta and inferior vein cava and perfuse for 60 minutes. This is an ideal model for pathophysiological research of isolated perfusion.
