摘 要:采用液相色谱法分离、蒸发光散射检测器检测,外标法定量测定牛乳中乳果糖的含量。此法是将样品通过乙醇沉淀蛋白后滤液上机测定,较酶法具有快速、准确、操作简单、易于掌握等特点。其色谱条件为:色谱柱HPX-87P,300 mm×7.8 mm;流动相(乙腈∶水)为257∶5;流速1.00 mL/min;柱温60℃。此方法的回收率为97.92%;CV为2.22%。The lactulose in the sterilization milk was formed a isomer in the higher temperature, which could be scaled the degree of head in milk. The test adopted HPLC to separate; ELSD to detect and external tap to fix quantity to determine the content of lactulose. The process was: the filtrate that the sample was been deposited protein with ethanol was detected, this measure was faster, accurater, easily operated and masteried than the way of enzyme. The condition of chromatogram was :the columm was HPX-87P, 300×7.8mm; mobile phase(ACN: water) was 25:75; the velocity of flow was 1.00 ml/min; the column temperature was 60 ℃. The recovery was 97.92%; CV% was 2.22.
