摘  要:对枯草芽孢杆菌胞外蛋白酶的产生条件及其培养液水解鱼蛋白质制备蛋白胨进行了初步研究.结果表明,适宜枯草芽孢杆菌产蛋白酶的培养基初始pH值7.0、培养温度30℃、250 mL三角瓶中的装液量为60 mL,培养时间为24 h;用枯草芽孢杆菌培养液为蛋白酶源水解蛋白质时,应控制反应体系的温度为55℃,pH 9.0,加入低浓度的Mg2+、Ca2+并去除Cu2+、Fe3+、Pb2+和Ag+,对枯草芽孢杆菌产酶有促进作用.用枯草芽孢杆菌培养液分别水解鳕鱼粉和罗非鱼,蛋白质制备蛋白胨的得率分别为13.95%和14.96%,与目前的工业水平基本相当.Proteinase activity, cuhure conditions, hydrolysis conditions and peptone recovery rate were studied for the feasibility of producing peptone by hydrolyzing fish protein with Bacillus subtills culture. It was strongly indicated that Bacillus subtilis cultures had typical character of proteinase and fish protein could be effectively hydrolyzed by the promising proteinase resource. It was also indicated that the initial media pH 7.0, culture temperature of 30 ℃, of 60 mL meding in 250 mL flask and fermentation time of 24 h were suitable for obtaining Bacillus subtilis cultures with high proteinase activity. It may be seen that controlling hydrolysis temperature at 55℃ and hydrolysis pH at 9.0, adding Mg^2 + and Ca^2 + at low concentration and removing any Cu^2+ , Fe^3+ , Pb^2+ and Ag^+ were good means to keep high proteinase activity when protein was hydrolyzed using Bacillus subtilis cultures as proteinase resource. Recovery rates of peptone thus obtained were 13.95 % and 14.96 % for Tilapia meal and fishmeal of codfish, respectively, which were corresponding to the current industrial level.
