树莓超氧化物歧化酶的分离纯化和部分性质研究  被引量:10

摘  要:树莓SOD粗酶液经硫酸铵分级沉淀、ScphadcxG-100柱层析,同时对比DEAE-52柱层析和金属螫合亲和柱层析,并对其部分性质做了研究。从树莓中纯化得到3种Mn-SOD同工酶:SOD Ⅰ、SOD Ⅱ、SODⅢ。结果表明经金属螫合亲和柱层析的回收率和纯化倍数远高于DEAE-52柱层析。3种SOD对KCN和H2O2均不敏感,最大紫外吸收峰均为280nm,SOD Ⅰ和SODⅡ受温度影响较小.SODⅢ受温度影响稍大,pH值在7~9范围内3种SOD的相对酶活力较高。SOD Ⅰ ,SODⅡ and SODⅢ were tration on SephadexG-100 and comparing purified from the raspberries by ammonium-sulfate, gel fil DEAE 52 on exchange chromatography with Metals Chela ring AffinityChromatogram. And some of its characters were analyzed.The results showed that the specific activity and the recovery of the three enzymes from Metals Chelating Affinity Chromatogram were superior to that from DEAE- 52 on exchange chromatography . All the three enzymes were not sensitive to KCN and H2O2. And the uhraviolet absorption peaks value of them were all 280nm. The effect of temperature on SODⅠand SODⅡwere small, but that on SODⅢ was bigger. The relative activity of SOD were higher when pH value was in range between 7 and 9.
