摘  要:目的:观察榄香烯对小鼠肝细胞癌的治疗作用,为临床肝癌的治疗提供理论依据。方法:做荷瘤小鼠动物模型,癌瘤内多点多次注射给药,用RT-PCR方法测其多药耐药基因(multidrugs resistancegene,mdr1)的表达,同时观察抑瘤率、肿瘤坏死情况和小鼠死亡率。结果:荷瘤组(LT)、化疗组(C)、榄香烯组(E)和化疗药+榄香烯组(CE)的mdr1分别为(92·9%、100·0%、14·3%和14·3%),其中E、CE与LT的mdr1比较,P<0·05;LT++(42·9%)与C++(85·7%)的mdr1比较P<0·05。C、E和CE抑瘤率分别为(35%、27·2%和41·8%)与LT比较P<0·05。C、E和CE的死亡率分别为(15%、10%和5%)与LT(40%)比较P<0·05。病理示CE肿瘤坏死明显,大体见肿瘤腐肉状,色晦暗,质软,小鼠生存活跃。结论:瘤株Hca-A2/p的mdr1的自然表达率为92·9%。化疗使mdr1强表达增加。榄香烯可降低mdr1的表达并有良好的抗肿瘤的作用,与化疗药合用抗肿瘤作用更佳。OBJECTIVE:To offer basic theory for clinical liver carcinoma treatment by observing the therapeutic effects of Elemene in loading tumor mice. METHODS: The pharmaceuticals were injected into tumor of loading tumor mice in different points for many times, and then, determining expression of multidrugs resistance gene (mdr1) by RT-PCR, noticing inhibition rate of tumor, pathol- ogy of tumor and of mortality mice. RESULTS: Expressions of mdrl in loading turner group (LT), cbemiotberapy group (C), Elemene group (E), cbemiotberapy + Elemene group (CE) were 92.9 %, 100.0 %, 14.3%, 14.3 %, respectively and there was a statistical significance between E (or CE) and LT, P〈0.05, between LT ++ and C ++, P〈0. 05. Inhibition rates of tumor of C,E, CE were 35% ,27.2% ,41.8%, respectively and there was a statistical significance, compared with LT, P〈0.05. The mouse's mortalities of C, E, CE were (15%, 10%, 5%) respectively and there was a statistical significance, compared with LT(40%), P〈0. 05. The tumor's necrosis was outstanding and the mice were more energetic than others in CE. CONCLUSIONS.. The innate expression of mdrl of Hca-A2/p is 92.9%. The over expression of mdrl of Hca- A2/p increased with chemotherapy. Elemene can reverse mdrl gene and it is effective in killing tumor by itself and more effective by combining it with chemotherapeutic agents.
