摘  要:测定分析纯化的香菇多糖的单糖组分。单糖经α-萘胺衍生化以后,用硼砂做电泳介质,实现高效毛细管电泳分离,得到标准的α-萘胺衍生单糖的毛细管区带电泳谱图。将香菇多糖水解成单糖,用相同的方法实施电泳,得到多糖的毛细管区带电泳谱图。最佳工作条件:运行缓冲液为75 mmol/L、pH10.5的硼砂水溶液,分离电压15 kV,进样压力0.5 Psi,进样时间15 s,柱温25℃。通过与标准谱图对照分析,香菇多糖由鼠李糖、木糖、葡萄糖、甘露糖、阿拉伯糖和半乳糖组成。高效毛细管电泳能有效分析多糖中单糖组分,灵敏度高,分离效果好。The monosaccharide components in purified mushroom polysaccharides were analyzed.The monosaccharides were derived with α - naphthylamine, and separated by high - performance capillary electrophoresis with the borax buffer solution as medlum.Then the standard dectrophoretogram of a - naphthylamine derived monosaccharides was got. Polysaccharides from mushroom were hydrolyzed into monosaccharides and electrophoresised by the same method, so the electrophoretosram of polysaceharide was got.The best working condition were as follow: running buffer solution was 75 mmol/L, borax solution with pi110.5, running voltage was 15 kV,injection pressure was 0.5 Psi,injection time was 15s,column temperature was 25℃ .Compared with standard dectrophoretogram, the mushroom polysaccharides were composed of rhamnose, xylose, glucose, mannose, arabinose, galactose. The monosaccharide components of polysaccharide could be analyzed by HPCE effectively with high sensitivity.
