摘 要:目的比较两种外周血淋巴细胞染色体G显带方法的实际效果。方法制备人外周血淋巴细胞中期染色体标本,分别用GTG法和EDTA法进行处理后利用染色体自动分析系统观察,比较两种显带方法的利弊。结果GTG法深浅带带纹反差明显,但易出现显带不全、消化过头、分散度降低等问题。EDTA法不改变染色体分散度,不存在消化过头的问题,可利用核型多,可部分克服GTG法的不足,但此法所示带纹深浅带间反差不如GTG法明显。结论EDTA法显带染色体能基本满足辩识的要求,稳定性和可重复性较好,但在方法学上有进一步探索的必要,以提高染色体深浅带带纹反差。Objective : To compare the real effects of two methods for chromosome G-banding of peripheral blood lymphocyte. Methods: Metaphase chromosomes of peripheral blood lymphocyte were prepared and chromosome banding were performed by GTG ( G-band by Trysin using Giemsa, GTG ) method and EDTA (ethylenediamine tetraacetic acid, EDTA) solution, respectively, and their advantages and disadvantages were discussed. Results:The sharpness of chromosome bands induced by GTG method was good, but GTG method was prone to resulting in problems of incomplete banding or over digestion and decreasing chromosome dispersity, etc. Preferabaly, however, EDTA solution did not chang the chromosome dispersity and had an increased number of analysable metaphases, without problems of over digestion and decreasing chromosome dispersity, which partially overcame the shortages of GTG method. Conclusion : Chromosome banding by EDTA method can meet the routine need for chromosomes discrimination with better constancy and reproducibility, but it is much needed to further improve its effect on enhancing the contrast of dark and light bands of chromosome.
