摘 要:目的:PP1195是新近获得的一个基因,酵母双杂交发现PP1195与纤粘连蛋白(Fibronectin,FN)相互作用。本文旨在说明该基因与FN间的关系及对肝癌细胞浸润和迁移能力的影响。方法:采用体外翻译表达PP1195基因和FN基因产物,体外结合实验观察两者之间的作用;采用细胞粘附实验观察PP1195基因表达对Hep3B细胞与FN结合能力的影响;采用细胞侵袭重建基底膜和划痕实验观察该基因表达对肝癌细胞浸润和迁移能力的影响。结果:表明PP1195和FN有相互结合作用;诱导PP1195基因表达可显著增加Hep3B细胞与FN的结合能力;以及SMMC7721细胞的浸润和迁移能力。结论:PP1195基因是一个新的与纤粘连蛋白相互作用并能够增加肝癌细胞浸润和迁移能力的基因。Objective: To investigate the function of the PP1195 protein in hepatocarcinoma cell lines. Methods: In vitro translation was applied to express PP1195 and fibronectin (FN) gene products, and then an in vitro binding experiment was used to observe the interaction between the two proteins. A cell adhesion experiment was used to observe the effect of the expression of the PP1195 gene on the binding capacity for FN of Hep3B cells and Hep3B cells overexpressing PPl195. The cells were observed for their invasion potential after scoring the bottom of the fibronectin-coated flask. Results: We have shown that there was an interaction between the PP1195 protein and FN. Induction of the expression of PP1195 can significantly improve the binding capacity of Hep3B cells to FN. The infiltration and migration potentials of SMMC7721cells were also significantly increased. Conclusion: PP1195 is a novel gene whose product can interact with fibronectin and can increase invasion and migration in hepatocarcinoma cells.
