摘 要:目的:观察中药髓复康(SFK)对实验性脑脊髓损伤大鼠血液流变性的影响。方法:雄性SD大鼠80只,通过脑缺血再灌注和脊髓半横断损伤实验,观察SFK对于脑水肿和脊髓损伤后血液流变性和基因表达谱的影响。结果:①脑缺血再灌注损伤后,大鼠全脑含水量明显增加,SFK干预后仅轻微增加;②脑脊髓损伤后血液由低凝转为高凝,SFK干预后使高凝转为低凝;③基因芯片检测发现,脊髓损伤后出现众多的差异表达基因,SFK干预可以调节某些基因表达的,如上调脂皮素、载脂蛋白CI-II、半胱氨酸蛋白酶和纤溶酶原,下调弹性蛋白酶和血管内皮粘附分子。结论:SFK能纠正脑脊髓损伤早期的血液流变性和基因表达异常。Objective: To observe the influence of SFK (Traditional Chinese Medicine) on hemorheological change induced by cerebral and spinal cord injury. Method: Through brain Ischemic-Reperfusion and half-transection spinal cord injury,observe the effects of SFK on hemorheological changes and spectrum of gene expression.Results: Cerebral water content of rats in group of I/R model was evidently increased compared with that of rats in normal control and ASCI group.② After injury the hemorheological property was changed from lower coagulability to higher coagulability during the time from 8~ th hour to 24~ th hour.After admimstrated with SFK, the hemorheological property was changed from higher coagulability to lower coagulability. ③ With gene chip technique,various differential expressing genes were found. After administrated with SFK, expressions of LC-1, ApoCIII, CtsK and Plg were up-regulated and expressions of Ela and VCAM were down-regulated.Conclusion: SFK could not only effectively reverse the abnormal hemorheological status into normal one, but also regulate the expression of related genes.
