摘 要:探讨了雨生红球藻(Haematoccuspluvialis)712株的适宜培养条件及藻体诱导累积虾青素的培养基条件。重点研究了温度、pH和光照条件对雨生红球藻营养生长的影响,以及NaNO3、Fe2+盐和乙酸钠浓度对雨生红球藻诱导累积虾青素含量的影响。结果表明:24℃、1000~1500lx连续光照、pH8.0左右的生长条件适合雨生红球藻游动细胞增殖,使平均生长速率达到0.252/d。通过正交试验表明:缺氮培养基对于雨生红球藻细胞累积虾青素最为有利,虾青素含量达到6.72μg/mL,而FeSO4和乙酸钠浓度对虾青素的累积无显著性影响。Optimal culture conditions of the Strain 712 of Haematoccus pluvialis and the media condition to the accumulation of astaxanthin were discussed in this paper. The effects of temperature, pH and illumination conditions on the culture of Haematoccus pluvialis was studied. And the effects of the concentration of NaNO3, FeSO4 and sodium acetate on the accumulation of astaxanthin was also studied. The results suggested that the conditions such as 24 ℃, illuminated continuously under the intensity of 1 000-1 500 Ix, and pH 8.0 are suitable for the growth of mobile cells and the average growth rate was to 0.252/d. By orthogonal test, it suggested that media without nitric salts are suitable for the accumulation of astaxanthin, the astaxanthin content reached to 6.72 μg/mL, and concentrations of FeSO4 and sodium acetate have no significant effects on the accumulation of astaxanthin.
