摘 要:[目的]探讨牛磺酸抗电离辐射损伤机制。[方法]将ICR小鼠随机分为5组,其中3个实验组分别经口给予牛磺酸50,100,300mg/kg,共30d,辐照对照和正常对照组给予去离子水。第31天,除正常对照组外各组小鼠分别接受5GYCoγ射线一次性全身照射,观察牛磺酸对损害的保护效应及对抗氧化及免疫功能的影响。[结果]60辐照小鼠骨髓细胞微核率为26.6‰,显著高于正常对照微核率1.2‰,外周血白细胞数照射后3d下降为1.95×109/L,14d为3.80×109/L,明显低于辐照前7.60×109/L(P<0.05),骨髓细胞DNA含量也显著下降(P<0.05)。50mg/kg牛磺酸组,血清SOD活力为110.4U/ml,显著高于辐照对照组84.3U/ml(P<0.05);100mg/kg牛磺酸组,SOD活力、MDA含量、溶血素抗体积数明显优于辐照对照组;300mg/kg牛磺酸组SOD为137.8U/ml,MDA含量为5.8nmol/ml,溶血素抗体积数为83.8,脾淋巴细胞增殖试验光密度差值为0.154,外周血白细胞总数及骨髓细胞DNA含量上升,微核率下降,30d存活率明显提高,与辐照对照组差异有统计学意义。[结论]牛磺酸能够改善Coγ辐射对小鼠60损伤的生物学效应,这与其提高抗氧化能力及增强免疫功能有关。[ Objective ] To study the protective mechanism of taurine against damage induced by ^60Coγ -ray irradiation. [ Methods ] Mice were fed with taurine at dose of 50 mg, 100 mg, 300 mg/kg for 30 days, positive ( irradiation ) and negative control were administered with distilled water only, then all mice except negative control group were exposed to 5,0 Gy T -ray for whole-body-irradiation at the 31st day. Observe the protective effect of taurine against damage of irradiation and it' s effects on antioxidant system and immune function. [ Results ] The bone marrow micronuclei rate of positive ( irradiation ) control group increased to 26.6‰, significantly higher than 1.2%o for negative control. Total amount of periphery WBC decreased to 1.95 ×10^9/L, 3.80 ×10^9/L respectively at 3rd and 14th day post-irradiation, significantly lower than pre-irradiation; DNA content of bone marrow cell also remarkably decreased ( P 〈 0.05 ) . In test groups, activity of serum superoxide dismutase ( SOD ) elevated to 110.4 U/ml in 50 mg/kg taurine group, significantly higher than positive control; SOD activity, MDA content and serum hemolysin were significantly better in 100 mg/kg taurine group than positive control; in 300 mg/kg taurine group, total amount of periphery WBC and DNA content increased, bone marrow micronuclei rate decreased, SOD activity elevated to 137.8 U/ml, MDA content decreased to 5.8 nmol/ml, serum hemolysin enhanced to 83.8, OD value for lymphocyte proliferation improved to 0.154, and 30 day survival rate also enhanced. Those changes were all significant compared with positive control ( P 〈 0.05 ) . [ Conclusion ] Our result suggests that taurine can alleviate irradiation damage induced by gamma-ray, might be associated with ameliorating antioxidant level and immune function.
