摘 要:目的:观察应用药物缓释剂和卡介苗构建大鼠局限性炎性肉芽肿模型的效果。方法:正常Wistar大鼠36只,随机分成3个对照组和3个实验组。对照组每组4只,注射药物分别为0.1mmol/LPBS(对照1组)、10g/L明胶(对照2组)和体积分数为25%的泊洛沙姆407(P407)(对照3组)。实验组每组8只,注射药物分别为卡介苗(BCG)(BCG组)、BCG-明胶(明胶组)和BCG-P407(P407组)。开腹直视下肝内和皮下分别注射药物0.1ml。术后4周、8周时进行形态学观察和分析。结果:各对照组肝脏和皮下均未见明显变化。术后8周时,BCG组肝脏和皮下结节已经明显发生纤维化;明胶组和P407组仅见少量纤维组织增生,但明胶组有腹腔粘连发生。结论:应用BCG-P407可在大鼠肝内和皮下建立持续的局限性炎性肉芽肿模型。Aim : To explore the effects of granulomatous inflammation model in rats established by the application of drug sustained release and Bacillus Calmette-Guerin (BCG). Methods: A total of 36 Wistar rats were randomly divided into 3 control groups and 3 model groups. There were 4 rats in each control groups and 8 rats in each experimental groups. PBS at 0.1 mmol/L, gelatin at 10 g/L, 25% Poloxamer 407, BCG, BCG-gelatin, and BCG-Poloxamer 407 were respectively injected into liver and subcutaneous tissue of the rats in control groups and model groups. The pathological changes were observed macroscopically and microscopically at 4 weeks and 8 weeks after injection. Results: No distinct changes were observed in control groups. Eight weeks after injection, serious fibrosis was observed in BCG group, and only a little fibrous tissue was observed in BCG-gelatin group and BCG-P407 group. But abdominal adhesion could be seen in BCG-gelatin group. Conclusion : Steady localized granulomatous inflammation could be successfully established in liver and subcutaneous tissue in rats by BCG-P407 without significant systemic influence on experimental animals.
