摘 要:目的:研制保健食品中绿原酸的单扫描极谱分析方法。方法:样品经50%的乙醇溶液超声振荡提取,用拟定的极谱方法进行定性和定量测定。结果:在0.05mol/L硫酸、3mol/L亚硝酸钠、2.5mol/L乙酸钠介质中,绿原酸峰电位为-172mV(vs.SCE)。绿原酸浓度在5.0-50.0μg之间线性关系良好,相关系数r=0.9993,回归方程Y:2.6923X-2.365。相对标准偏差(RSD)为1.70%~6.66%(n=5)。加标回收率为94.3%~97.1%(n=9)。结论:建立的单扫描极谱法测定绿原酸的方法简便快速、结果准确、灵敏度高、仪器廉价、易于普及。适于实验室检测样品中绿原酸的含量。Objective: Single - sweeping polarograph for the analysis method of chlorogenic acid in health foods was studied. Methods :The chlorogenic acid was determined by the method qualitatively and quantitatively. Results:In the supporting electrolyte solution that composed of 0. 05 mol/L sulfuric acids, 3 mol/L sodium nitrite, 2.5 mol/L sodium acetate, the peak potential of chlorogenic acid was - 172 mV( vs. SCE). The peak current of chlorogenic acid was linear to the concentration of chlorogenic acid in the range of 5.0 - 50. 0 μg. The coefficient of correlation was r = 0. 9993. The regression equation was Y = 2. 6923X -2. 365. Its relative standard deviation (RSD) was in the range of 1.70% -6. 66% (n = 5). Recovery of chlorogenic acid was in the range of 94. 3% - 97. 1% ( n = 9). Conclusion : The method of single - sweeping polarograph for the determination of chlorogenic acid is simple, rapid and the results are accurate and highly sensitive. The single - sweeping polarograph is cheap and easy to popularize. The method can be applied to the determination of chlorogenic acid in lab.
