摘 要:目的观察维生素E(VE)补充对大鼠肝线粒体ATP酶和抗氧化酶活性的影响。方法Wistar大鼠随机分成4组,对照组、VE1、VE2和VE3干预组;对照组给予普通饲料,3个干预组均喂饲添加维生素E的饲料,剂量分别为335,1 340,5 025 mg/kg饲料,喂养10周后,摘取肝脏提取线粒体,测定Na+-K+-ATP酶、Ca2+-Mg2+-ATP酶、谷胱甘肽过氧化物酶(GSH-Px)、超氧化物歧化酶(SOD)的活性及丙二醛(MDA)的含量。结果VE1组与对照组相比,SOD、GSH-Px、Na+-K+-ATP酶、Ca2+-Mg2+-ATP酶活性显著升高(P<0.05),MDA水平显著降低(P<0.05)。VE2、VE3组与对照组相比未见改善。VE2、VE3和VE1组相比,SOD、GSH-Px、Na+-K+-ATP酶、Ca2+-Mg2+-ATP酶活性显著降低(P<0.05),MDA水平显著升高(P<0.05)。结论补充适量VE(335 mg/kg)能显著增强大鼠肝线粒体ATP酶活性及氧化能力;较高剂量VE;(1340,5 025 mg/kg)组未能改善大鼠肝线粒体ATP酶活性及抗氧化能力。Objective To study the effect of vitamin E(VE)on antioxidative enzyme and ATPase activity of hepar mitochondria in rats. Methods 40 Wistar rats were randomly divided into 4 groups based on body weight, which was control, VE1, VE2, and VE3 group. The control was fed with normal rat diet and the other three interfered groups were supplemented with different levels of VE enriched diets which contained VE 335, 1 340, 5025 mg/kg, respecively. The perido of the trial was about 10 weeks. At the end of the trial, the mitochondric were extracted from hepars. The activties Na^+ -K^+ -ATP ase, Ca2^+ -Mg2^+ -ATP ase; Superoxide dismutase(SOD) ; Glutathione peroxidase( GSH - Px) and the contents of malondialdehyde (MDA)were measured. Results Compared with the control group, VE1 group increased the activities of Na^+ -K^+-ATP ase, Ca2^+-Mg2^+ -ATP ase, SOD, GSH- Px(P 〈 0.05 ), and decreased the contents of MDA( P 〈 0.05 )o Compared with the control group, the activities of antioxidant enzymes and ATPase in the hepar mitochondria of VE2 ; VE3 group had not much change. Compared with VE1 group, VE2; VE3 group decreased the activities of Na^+-K^+ -ATP ase, Ca2^+-Mg2^+ -ATP ase, SOD, GSH- Px(P〈0.05),increased the contents of MDA(P〈0.05).Conclusion 335 mg/kg VE could increase the activities of antioxidant enzymes and ATPase. 1 340, 5 025 mg/kg VE could not further increase the activities of antioxidant enzymes and ATPase.
