摘  要:目的以聚氰基丙烯酸丁酯纳米粒(PBCA-NP s)作为端粒酶逆转录酶(hTERT)反义寡核苷酸(A-SODN)载体,转染肺癌A 549细胞,观察其对该细胞周期和凋亡的影响。方法PBCA-NP s携带hTERT A SODN转染A 549细胞后,绘制细胞生长曲线表达细胞生长状态的变化;流式细胞仪(FCM)检测细胞周期及细胞凋亡情况。结果A SODN转染后A 549细胞增殖减慢;FCM检测结果显示:A SODN转染后细胞周期变化明显,细胞被阻滞于G0/G1期,细胞凋亡率明显增加。结论hTERT A SODN能有效改变A 549细胞的细胞周期,诱导细胞凋亡的发生。[Objective] To investigate the effect of hTERT antisense oligodeoxynucleotide on cell cycles and apoptosis of A549 cells mediated polybutylcyanoacrylate nanoparticles (PBCA-NPs). [Methods] Cell proliferation after transfecting hTERT ASODN was observed by cell growth curve; cell cycles and apoptosis rate were analyzed by flow cytometry (FCM). [Results] Cell proliferation was inhibitted after being treated with hTERT ASODN; and cell cycles were blocked in G0/G1 phase,cell apoptosis rate increased obviously. [Conclusion] hTERT ASODN mediated by PBCA-NPs can change A549 cell cycles and induce the cell apoptosis efficiently.
